Rational preparation of powder anthocyanin incorporated-urease test for detection of K. pneumoniae and P.mirabilis

In this present study, we developed Brassica oleracea var. Capitata (red cabbage) extract (RCE) powder- incorporated colorimetric media for rapid, selective, sensitive detection of urease-postive bacteria, Klebsiella pneumoniae ATCC 54475 and Proteus mirabilis 25933. The major component RCE is anthocyanin groups which are in charge pH dependent color change. ureas test contains certain concetration anthocyanin, urea prepated 0.01 M sodium phosphate buffer azide. the was hydrolyzed by urease enzyme released from K. P.mirabilis produce ammonium (NH3) molecules sequentially made alkaline, caused deprotonation induced change pink to green. Also, Escherichia coli 25922 used a urease-negative control bacteria. P. examined as function concentration incubation time. lyophilized powder form discard interference free proton liquid owing addition hydrochloric acid (HCl). We also performed digital image analysis, Delta E (∆E) quantitative confirmation visual results. It observed that results showed green 150 210 minutes incubaiton mirabilis, respectively. Here in, successfully prepared anthocyanin-incorporated perform with naked eyes analysis.
